Circadian Clock Controls Insulin and Blood Sugar in Pancreas
The circadian clock is a molecular mechanism that regulates physiological responses of an organism to changes in the daily light cycle. Disruptions to the circadian clock have been linked to metabolic disorders such as cardiovascular disease, obesity, and type 2 diabetes. However, the exact mechanism underlying these disorders is poorly understood. Joseph Bass, a member of a CBC 2007 Catalyst team, is senior author on a recent Science paper, "Pancreatic β cell enhancers regulate rhythmic transcription of genes controlling insulin secretion," which provides new insights into how circadian regulatory processes impact pancreatic function. The work is a collaborative effort including several researchers from NU and UChicago. (read full story)

CBC Awards Help Move Promising Synthetic Antibody Technology into the Commercialization Pipeline
A 2009 CBC Spark Award to Anthony Kossiakoff (UChicago) and Vladimir Gelfand (NU) allowed the development of a high-throughput technology platform to efficiently produce customized, engineered, “synthetic antibodies.” Advances made with Spark funding allowed Kossiakoff and colleagues to win a $12 million center grant from the NIH, which was matched with a $2.3 million Lever Award from the CBC in 2012. In July 2015, the pharma company Celgene committed to a 3 year, $25 million partnership to use the technology to develop next-generation cancer drugs based on synthetic antibodies. (more info)

CBC Funding Contributes to Understanding of the Gut Microbiome’s Role in Infants with Allergies to Cow’s Milk
According to a new study published on September 22, 2015, in The ISME Journal, “Lactobacillus rhamnosus GG-supplemented formula expands butyrate-producing bacterial strains in food allergic infants,” the gut bacteria of babies who developed tolerance to cow’s milk after treatment with probiotic formula was significantly different from those who remained allergic. The study points out that identification of bacteria that can protect against the development of food allergies could jump-start the search for novel therapeutics. Aly Azeem Khan, a co-author on the publication, was supported by the CCSB/CBC Fellows Program, financed by the first CBC Lever Award to the Chicago Center for Systems Biology (CCSB). (more info)

Synaptic Membrane Nanodiscs May Help Find Alzheimer’s Disease Therapeutics
Congratulations to William Klein of NU for his recent publication in PLoS One: “Nanoscale Synaptic Membrane Mimetic Allows Unbiased High Throughput Screen that Targets Binding Sites for Alzheimer’s-Associated Aβ Oligomers.” Using a High Throughput Screening (HTS) facility at NU, Klein and colleagues developed a cell-free system consisting of a library of synaptic proteins individually embedded in nanoscale lipid bilayers (Nanodiscs). Nanodiscs proved successful in a pilot HTS screen for small molecule inhibitors of amyloid β oligomer (AβO) binding. This novel technique appears promising for future large-scale HTS screens for membrane protein targets, including potential Alzheimer’s disease therapeutics. The publication is attributed to a CBC HTS Supplemental Grant awarded to Klein in June 2013. (more info)
